Abstract

The spectlnomycin resistant strata of slow growing R. japonkum R-168 was selected to

be participated In conjugation with 1~ call WA003/pGS9. Tay was introduced from suicide vector pGS9 Into R. japonkum R-168 spr chromosome at the frequency of 1.0 x 10 4- 5.0 x 10¢¥ 7 and the tramconjngants were selected on the yeast e:Uad-ranaltol plate containing kananjda (58 g/mo and specdoomyda (100pg/aO after 0-9 days incubation. AN traasconjaaats we tested were fumed to coutata To S DNA on their geuome, which was confirmed by Southern hybridizat on experineats. R. japonkum RMa75, which had been sekded through plant test, was found to be defective In symblodc nitrogen Mug ability and the production of kgbemoglobin in soybean nodules formed by the inoculalion of this mutant. In addition, this mutant strain hardly developed m1h agenase activity nymbiodaiy In contrast with the wild type strain, Indicating that some nitrogen fixing gene might be blocked in this strain and the production of leghemoglobi could be decreased by the Inkrkssoe in sttrugrsi fhdng genes.
